Citrate Synthase Overexpression of <i>Pseudomonas putida</i> Increases Succinate Production from Acetate in Microaerobic Cultivation

Acetate is an end-product of anaerobic biodegradation and one the major metabolites microbial fermentation lingo-cellulosic hydrolysate. Recently, acetate has been highlighted as a feedstock to produce value-added chemicals. This study examined conversion succinate by citrate synthase (gltA)-overexpressed Pseudomonas putida under microaerobic conditions. The metabolism initiated with gltA enzyme, which converts acetyl-CoA citrate. gltA-overexpressing P. (gltA-KT) showed ∼50% improvement in production compared wild type. Effect of overexpression of Actinobacillus succinogenes phosphoenolpyruvate carboxykinase on succinate production in Escherichia coli.

Succinate fermentation was investigated in Escherichia coli strains overexpressing Actinobacillus succinogenes phosphoenolpyruvate carboxykinase (PEPCK). In E. coli K-12, PEPCK overexpression had no effect on succinate fermentation.
